BIO 221 - Molecular Biotechniques

4 credit hours - Three hours of lecture and three hours of laboratory weekly; one term.
Investigate the theory and application of molecular biology as they apply to research, medicine, agriculture, bioremediation and criminology. Use extensive hands-on procedures in the integrated lecture/lab to reinforce the theory behind cloning, genetic engineering and forensic DNA science. Learn concepts that govern the use of molecular techniques and the applicable laboratory skills. Lab fee $40.

Prerequisite(s): BIO 101  or permission of department chair and eligibility for ENG 101 /ENG 101A  and any general education math.

Note: Typically offered at MC; spring term.

Course Outcomes:
 

  1. Perform calculations and procedures to make solutions of specific concentrations. 
  2. Generate a standard curve and use it to determine the concentration of an unknown solution.
  3. Describe the use of the Global Harmonized System to determine the hazards associated with various reagents, and describe the use of safety equipment and procedures associated with a molecular biology laboratory.
  4. Use aseptic technique in various procedures to culture microorganisms.
  5. Describe and perform procedures to isolate pure genomic and plasmid DNA from bacterial cultures.
  6. Conduct all steps in the polymerase chain reaction including primer design and preparation, amplification, and agarose gel electrophoresis.
  7. Conduct DNA analysis including restriction endonuclease analysis, determination of concentration, DNA sequencing reaction preparation and analysis.
  8. Utilize NCBI database resources to conduct literature searches, obtain DNA and protein sequence information, perform sequence comparisons and design primers.
  9. Explain the theory of Southern, Northern and Western hybridization techniques. 
  10. Troubleshoot laboratory experiments.
